How many days of rest does it take before you start loosing muscle due to lack of exercise?

A couple of days rest is optimal, you need to grow, recover and recouperate.
In theory you'll lose strength within about 36 hours. If you eat well and stay moderately active you wont lose too much mass. I've seen people off weights/out of the gym for months at a time and they dont lose much, and whatever they do lose comes back very quickly.

I think you actually start to loose muscle after 2 weeks not that your not going to get weaker in the 2 weeks your just not going to really start to lose muscle mass till then
in theory then once you get your body to where you want it as long as you can keep up a routine once a week you should allways be able to do it though fat might start to build up and weigh you down if your not doing something oftenly or watching your diet
