Well, according to BoxRec the answer is Peter Maher - who (not counting the initial 1st round ko on his record, which was an amateur bout) is credited with 48 knockouts in the 1st round!
However, many of these quick wins were against men, who had never fought before (and would never fight again!)... so they were probably more exhibition-style fights, where a local strong boy wanted to try his luck against a real pro fighter. I'm not sure, we should count these "fights" as real pro bouts.
Other boxers with very high numbers in this category (like Lamar Clark and Young Otto) have lots of fights like that - so their records should also be taken with a grain of salt, I think.
Shannon Briggs, on the other hand, has scored 33 1st round knockouts against oponents, who have all engaged in other pro bouts, besides those against himself. These fights should probably be counted as actual pro bouts.
So I would go with Shannon Briggs as the answer to this question.

Shannon Briggs was my pick @ 33. Otto and Maher do have more though and a few more Briggs 1st round wins will help him pass Otto.

Someone just asked me that question, and if you go BoxRec, like Bundana stated, it's Maher with at least 49, that is not counting the initial 1st round ko on his record, which was an amateur bout. Young Otto was always said to be the answer with 42, But BoxRec has him at 35.
After his recent 1st stoppage, I up-dated Shannon Briggs bio on BoxRec to read - Has 33 first-round knockouts, more than any other heavyweight champion in history. On at least seven occasions, Briggs has knocked out his opponent in less than 30 seconds.
