If Khan loses (which is unlikely at this point in his career) then his handlers will make plenty of excuses as to why he lost. They will demand an immediate rematch with the guy who beat him and he will get into the best shape he can and fight his best to try to take the guy out early to prove that the loss was bad luck.

If Khan loses, I don't think it'll affect his career any (in the long term at least). A lot of great fighters have early career losses, B-HOP is the first that comes to mind.
