The 28-year-old provided a boxing masterclass to see off the American on a unanimous points decision 119-109, 118-110, and 120-108.
Khan has repeatedly called out Mayweather, who is undefeated in 47 fights, but the 'Money' insists he wants to fight the Filipino Pacquiao first.
Last week Eddie Hearn said a fight between Brook and Khan at Wembley could be on the cards but the Bolton boxer insists he wants Mayweather first.
