It's easier to tell if someone is on roids by his hands, than his muscule mass. Muscle can be built. Hands are the size they are, if you go by the bone structure in his hands.
Manny was never as small as he fought, he did start low however. He drained himself. Hence the reason he was so easily stopped earlier in his career. It's easy to imagine him draining himself, when Freddie Roach has to literally stop him from overtraining. He was also a growing fighter, and started as a teenager.
Take Julio Cesar Chavez Jr. as of recent times. He started out at 130. This was only 6 years ago, and he's 'successfully' moved up about 25 pounds. Now, if only he had major skills that have been displayed on a more consistent basis.
Nonetheless, if you look at the size of a fighters hands, you can get a better feel for where he could be confortable, weight-wise. Manny has bigger hands than Cotto, who is a natural Welterweight.

Great example. I heard JCC Jr. doesn't really like to train either, so he could have probably stayed at welterweight a bit longer if he would have had Pacquiao's work ethic. The most notable thing Pacquiao and JCC Jr. have in common other than moving up through so many weights is the huge head. Both have huge noggins. Chavez-Vanda looked like a 5 pound medicine ball vs. a 20 pounder.
