I noticed the poll and the thread title have seperate questions. Being the best is one thing, and being the least likely to lose would be based on the level of opposition you're facing.

Don't know much about Calderon, I'm downloading his fights against Dieppa and Barrera now.

Calderon is probably better, but it is impossible to quantify. He's better than his competition than Cotto is anyway. Still, they are both elites.

Fres Oquendo is the only PR heavy with a pulse (though it is fluttering and extremely light, hah). Boxrec is listing Derrick Rossy unexpectedly as well. I learn something every day! Other potential ones you could have listed to fill out the options: Tito Trinidad (duh), Daniel Santos, Kermit Cintron, Juan Manuel Lopez, and uh... Well, there's crap like Carlos De Leon and Hector Camacho Jr. It is a small island. What they have is pretty incredible.
