I would have to lean towards the 7th game of the World Series.

It seems much more difficult to even get to a world series IMO.

Then after playing 100 plus games to fail during the crucial moment is enough to bring tears.

I say it's either a NCAA Championship. I'd roll with college football because you see the pain in these kids eyes as they come up short in the press conference. For most, that will be the closest they get to a professional level and to lose it in their final game is some of the most devastating footage I think I've seen. From a conference championship game or simply the national title game. NCAAF gets my vote

I thought it was a tossup between the Stanley Cup Finals, World Series & NBA Finals, because of the same reasons you mentioned. They play soo many games during the season and in the playoffs, and to be so close and lose it all by a field goal, a buzzer beater shot, or a goal out of nowhere has to deflate Pro athletes totally and completely.

I know it won't win because it isn't a popular sport but without a doubt in the the NHL Stanley Cup. All series are 7 game series and it is a very physical game. If you aren't a hockey fan just watch a few NHL playoff games and you'll understand how difficult it is to hoist the cup.

                  The World Cup final.

                  Four years of preparation.

I like this choice but part of me thinks the world cup is diminished by league play. I'm not a soccer honk but it seems the English fans, for example, care more about their city winning a championship than England winning a world cup. Could be the same thing in other nations too. I could be wrong though.
