When you're pushing you're using your lats/pecs/shoulders/quads/hamstrings/calves/triceps/biceps/forearms/abs etc.! You're using a huge amount of muscles.
Each play is like maxing out with all those muscles, it's extremely tiring especially considering you're trying to block a 6'7 290lb defensive end like Julius Peppers or a ton of other monsters. Some guys are so powerful they get double teamed and still break out of the double team. Can you imagine the power it takes to bowl over 2 300lb men that can bench and squat over 500lbs each? They're not tired from running at all.
It's sort of like wrestling, when you see these MMA guys exhausted from rolling on the ground it's because of all the muscles they're using.
